The 29-year-old South African carded an 8-under 62 on the par-70 Royal Birkdale course in the third round, making him the first male player to ever shoot 62 in a major.
Grace's round, the lowest in the history of the majors, saw him move from four over par overnight to four-under.
A 63 had been recorded 10 times previously in the British Open, twice previous year at Royal Troon, when Phil Mickelson did it in the opening round only for eventual victor Henrik Stenson to match him on the Sunday.
By the turn in his third round, Spieth - Masters and US Open champion in 2015 - had picked up three birdies to move to nine under par, two clear of his playing partner and fellow American Matt Kuchar.

Prior to Branden Grace making golf history with his legendary round of 62, there had been 31 different times where a player had shot a 63 in a major championship.
On the front nine, Grace, a native of South Africa, shot his age - 29 - and he did not look back for the rest of his eight-under round.
"Talking about this can go on forever", Grace said.
Grace started the day 4 over and in a tie for 45th place and finished at 4 under and just two shots out of the lead held by Jordan Spieth, who hadn't teed off yet when Grace finished his round.
To make matters even more fantastic, Grace did this after shooting 74 the round before during some brutally bad weather.
He then drained more putts for birdies on 14 and 16 before the two-putt on 17 meant he needed just a par from the last - where up to that point there had been just one bogey all day.
Grace said he was trying to finish the round without a bogey and wanted to make one final birdie on the last hole. In the opening round of the 1980 U.S. Open, Jack Nicklaus missed a three-footer on the final hole for 62.
